Description

The MACSima Platform by Miltenyi Biotec is an integrated, automated system for high-plex spatial biology analysis. At its core is an epifluorescence microscope system equipped with six high-power excitation LEDs (with filters at 386/23 nm, 420/10 nm, 470/40 nm, 531/46 nm, 628/32 nm, and 725/40 nm). The platform utilizes MICS (MACSima Imaging Cyclic Staining) technology to enable multiplexed protein and RNA detection on the same tissue section. It supports the analysis of hundreds of protein markers and dozens of RNA targets within a single sample run. The system is compatible with all types of fixed samples, including FFPE, PFA, and acetone-fixed tissues and adherent cells, using dedicated sample carriers. A key component is the associated antibody portfolio, which includes hundreds of biomarkers validated for the technology, available as individual reagents or in pre-configured panels. For transcriptomics, RNAsky Detection Probes can be used alongside protein markers. Automated experiment execution encompasses staining, washing, and imaging steps. Data analysis is performed using the MACS iQ View Analysis Software, which provides tools for visualization and quantitative assessment, such as t-SNE/UMAP plots and distance mapping. The platform is presented as a complete workflow solution, encompassing the instrument, consumables, reagents, and software.
