EarlyTox™ Caspase-3/7 R110 Assay Kit

Description

The EarlyTox Caspase-3/7 R110 Assay Kit provides a homogeneous, single-reagent-addition method for the quantitative measurement of caspase-3 and caspase-7 activity in cell lysates, supporting apoptosis research. Its operation is based on a bis-peptide substrate, (Ac-DEVD)2-R110, which contains two DEVD peptide sequences linked to the fluorogenic molecule Rhodamine 110. When active caspase-3/7 enzymes are present, they hydrolyze the substrate, releasing the R110 dye and producing a measurable increase in green fluorescence, with optimal excitation at 490 nm and emission at 520 nm. The kit includes purified R110 for generating a standard curve, enabling quantitative assessment of enzymatic activity. A competitive inhibitor, Ac-DEVD-CHO, is supplied for use as a negative control. Two kit sizes are offered: an Explorer Kit with materials for approximately two 96- or 384-well microplates, and a Bulk Kit containing reagents for about ten plates. The assay is specifically designed for use with fluorescence microplate readers.
