Description

Applied Scientific Instrumentation's diSPIM is a light sheet microscopy system built on an inverted microscope platform. The setup utilizes two water immersion objectives, each positioned at a 45-degree angle above the sample stage. These objectives are mounted on piezo-electric positioners for accurate movement during the acquisition of optical section image stacks. An optional third objective can be placed below the stage to provide an alternative viewing angle. This dual-inverted architecture illuminates a thin plane of light within the specimen from one direction while fluorescence detection occurs from the perpendicular axis. The method enables rapid, high-resolution three-dimensional imaging of live or cleared specimens while minimizing light exposure and associated photodamage.
