Description
The DAWN instrument, from Wyatt Technology (a Waters company), is a multi-angle light scattering (MALS) detector used for the absolute characterization of macromolecules and nanoparticles. It measures scattered light intensity at 18 discrete angles using high-gain, high-dynamic-range photodiodes. The standard light source is a 658 nm laser with programmable power output from 10% to 100%; an optional 785 nm laser is available to minimize fluorescence interference from certain samples. The instrument determines absolute molar mass and size (radius of gyration) without the need for column calibration. An optional dynamic light scattering (DLS) module can be added to measure hydrodynamic radius. Its effective measurement range extends from 200 Daltons to 1,000 Megadaltons. It is routinely used in conjunction with separation techniques like size-exclusion chromatography (SEC-MALS) or field-flow fractionation (FFF-MALS). Operation and data analysis are conducted through the ASTRA software suite, and a touch-screen interface provides real-time diagnostics. Applications include the analysis of proteins, polymers, viral vectors, and lipid nanoparticles.
