Description

The Applied Biosystems GlobalFiler, GlobalFiler IQC, and GlobalFiler Express kits from Thermo Fisher Scientific are multiplex PCR amplification kits formulated for forensic human identification using short tandem repeat (STR) analysis. Designed to meet the demands of expanding international DNA databases, these kits consolidate a comprehensive set of commonly used loci into a single, 6-dye reaction. The multiplex includes all markers recommended by the CODIS Core Loci Working Group, plus additional loci prevalent in European databases, facilitating data sharing and comparison across jurisdictions. A key feature is the inclusion of 10 mini-STR loci with amplicons under 220 base pairs, which improves success rates with degraded DNA samples. The kits differ in processing time: the GlobalFiler Express Kit is optimized for a rapid workflow with a sample-to-result time of under two hours, while the standard GlobalFiler and GlobalFiler IQC kits have longer amplification cycles, typically requiring 6 to 9 hours. The GlobalFiler IQC kit includes an internal quality control (IQC) marker to monitor PCR inhibition.
